SOP for Operation and Cleaning of HPLC Pump

Purpose

To lay down a procedure for Operation and Cleaning of Operation and Cleaning of High-Performance Liquid Chromatography (HPLC) Pump (Make: Waters, 515 Pump).

Scope

The scope of this SOP is applicable for Operation and Cleaning of Operation and Cleaning of HPLC Pump (Make: Waters) in the Quality Control (QC) laboratory at PharmaSky Ltd.

Responsibility

QC Personnel: To operate and clean HPLC Pump.

QC Supervisor: To verify operation and cleaning of HPLC Pump.

QC Head: To ensure compliance to the procedure.

Procedure

Procedure for Operation of HPLC pump

• Ensure that all the cables and power cords of the pumps are connected properly.
• Switch “ON” the HPLC Pump. The display will show, 515 Pump, 100 μl, Ver 1.0
• After a few seconds, the display will show, Flow 1.000, Ready, 0.000 ml, – 25 psi
• Press “Menu” till purge appears on the screen.
• Turn the black valve anticlockwise to purge the mobile phase.
• Press the EDIT/ ENTER key. The display will show messages ‘Start PURGE’ and ‘Press ENTER to confirm’.
• Press the EDIT/ ENTER key to start purging.
• The system will start the countdown from 5 minutes to zero. The display will show, PURGE5:00, RUN, 5.000 ml, -25 psi.
• After purging close the valve by turning it clockwise.
• Press “Menu” till ‘Flow’ appears on screen.
• Press EDIT/ ENTER Key to change the flow rate. Press the Up “Δ” and Down “↓” arrow keys to set the required flow.
• Press “Menu” key to register the set flow.
• Record the operation carried out using the HPLC pump in respective format for recording.

Procedure for Cleaning of HPLC Pump

• Clean the HPLC pump with a dry clean cloth.
• Ensure proper cleaning of HPLC pump.
• Record the cleaning of the HPLC pump in the respective format for recording.

During every operation and cleaning.
