What is baseline correction in UV VIS spectrophotometer?
The correction subtracts the absorbance value at a specific wavelength from all wavelengths across the sample spectrum. The correction is used to account for the effect of instrument noise and light-scattering particulates in the sample that can cause an offset in the overall sample absorbance.
What is baseline flatness in spectrophotometer?
Baseline flatness – the instrument is autozeroed with nothing in the sample compartment across its working range and this is then measured. It may be measured at zero absorbance or at a specified absorbance value. Reproducibility (both in terms of wavelength and photometric accuracy).
